It depends on what kind of information you want to obtain with the reading.
The readings which are the easiest to relate to your overall control (such as measured by your HbA1c) are those taken before eating. If you are on oral meds, these will probably suffice to get you a good control.
If your goal is to "calibrate" your insulin intake, you need both the before, and after, readings. I would also recommend both before and after if you are trying to control on diet and exercise alone.
Readings taken at various times throughout the day, recorded, and averaged over long periods like 14 or 30 days are the way to go if you want the "total picture".
