Yes, even one day missed can allow your body to ovulate and you to get pregnant.

My daughter got pregnant 3 times while on birth control for various reasons. The first time she missed one dose and didn't think it would matter - it obviously did. The 2nd and 3rd times she was on an antibiotic and wasn't warned antibiotics can cause the pill not to be effective that cycle.

Also, for whatever reason pregnancy tests didn't tend to work on her until 2 weeks or so after her missed period. They had to confirm her 3rd pregnancy by ultrasound. Her doctor did the pregnancy test which came back negative, but she knew what it felt to be pregnant and insisted to him she was pregnant.
